摘要：

目的：通过两种不同方法大量分离乳猪肝细胞，观察分离肝细胞的活性和功能，探讨猪肝细胞的分离方法。方法：本实验以肝上下腔静脉作为体外乳肝细胞分离的灌注口，用倒置相差显微镜、血细胞计数板计数，台盼蓝拒染试验判断细胞活力，并用自动生化仪进行肝细胞功能的检测。结果：实验组分离的每克肝肝细胞数和肝细胞活性明显高于对照组，两者差异有统计学意义(P 0.05)。结论：本实验改进了胶原酶体外两步灌流法分离乳猪肝细胞，为肝细胞的大量分离提供了一定的理论和实验依据。 Objective To study the method of primary porcine hepatocyte isolation by observe the hepatocyte quantity、viability and function.Methods Porcine hepatocytes were isolated by two-step perfusion through the supra-hepatic inferior vena cava,counting the hepatocyte quantity,observing the hepatocyte viability,and testing the levels of AST、ALT、LDH、TBIL.Results There were significant difference in hepatocyte viability and hepatocyte quantity per gram between two groups(P0.05).Conclusion The experiment improved the method of porcine hepatocyte isolation,and provided some theoretical and experimental evidence.
